Labial salivary gland biopsy is a reliable test for the diagnosis of primary and secondary amyloidosis. a prospective clinical and immunohistologic study in 59 patients

Abstract: Objective. Reports of the detection of amyloidosis by labial salivary gland (LSG) biopsy have been mostly anecdotal. The aim of this study was to assess the value of this method in the diagnosis of amyloidosis.Methods. LSG biopsy tissues were studied with a combination method using Congo red stain and immunohistologic characterization using an antibody directed against the serum amyloid P (SAP) component. Electron microscopy was performed in all cases. In a prospective study, we evaluated 30 patients with biop… Show more

“…Amyloid deposits in LSG usually appeared in a periductal distribution on CV or CR stained sections. These features were also found by Delgado and Mosqueda (10) and Hachulla et al (11). In the present study, the use of anti-AA antibody was helpful for confirmation of AA amyloidosis, particularly in cases with scant deposits of this substance.…”

“…All LNs were free from cancer metastasis. The constitutive pattern of CCL21 expression was also investigated in noninflamed peripheral tissues, including two normal synovial tissues collected during lower limb amputation, two normal minor salivary glands obtained from individuals with suspected amyloidosis 28 (normal histology assessed by routine examination; Congo red-negative), one normal colon obtained during surgical resection for cancer at a minimum of 5 cm from the tumor (normal histology assessed by routine examination), and one normal skin sample obtained from mastectomy. All tissues were fixed in formalin and embedded in paraffin.…”

“…Biopsy of the minor LSG is a sensitive, easily performed, and reproducible technique for amyloidosis diagnosis (6). In our patient, no amyloid deposits were found in the biopsied tissues or in LSG.…”
