Lack of association between atopic eczema/dermatitis syndrome and polymorphisms in the promoter region of RANTES and regulatory region of MCP‐1

Abstract: In this cohort of Hungarian children there was no association between - 28G, and - 403A alleles in the RANTES promoter, - 2518G polymorphism in the distal regulatory region of the MCP-1 and AEDS, or allergy.

“…[5][6][7][8][9][10][11][12] Therefore, RANTES could be considered a gene generally predisposing to autoimmune disease. To test this hypothesis, we investigated RANTES SNPs rs2306630, rs2280788 and rs2107538 in a group of 310 CD patients.…”

“…This gene has not yet been investigated in the context of T1D, although it has been shown to be associated with other autoimmune diseases, such as atopic dermatitis, asthma, sarcoidosis, rheumatoid arthritis and multiple sclerosis. [5][6][7][8][9][10][11][12] RANTES belongs to the family of CC chemokines, which are involved in immunoregulatory and inflammatory processes owing to their ability to recruit, activate and co-stimulate T cells and monocytes. 13,14 In addition to the trafficking effect, RANTES, like other CC chemokines, plays an important role in co-stimulation of T-cell proliferation 15,16 and activation of the T cells localized in the inflammatory lesion.…”

“…In addition, the GG genotype correlated with asthma severity and the G allele was associated with increased blood eosinophil levels 20 . However, there was no association between MCP-1-2518 and atopic eczema/dermatitis syndrome or allergy in a cohort of Hungarian children 21 . In case of sarcoidosis, MCP-1-2518 does not play a substantial role in the disease susceptibility, but might be related to the recruitment of monocytes/macrophages to the alveolar spaces in Japanese patients with sarcoidosis 22 .…”

“…8 The activities of CCL5 are mediated through its binding to CCR1, CCR3 and CCR5. 9 Genetic evidence exists that links CCL5 with numerous human diseases, such as coronary artery disease, 10 multiple sclerosis, 11 rheumatoid arthritis, 12,13 asthma and atopy, [14][15][16][17] atopic eczema/dermatitis syndrome, atopic dermatitis, 18 hepatitis C virus infection, 19,20 heart graft rejection 21 and systemic lupus erythematosus. 22,23 …”
