Lack of expression of bundle-forming pili in some clinical isolates of enteropathogenicEscherichia coli(EPEC) is due to a conserved large deletion in thebfpoperon

Abstract: Enteropathogenic Escherichia coli (EPEC) produces a plasmid-encoded type IV pilus, called the bundle-forming pilus (BFP), involved in the formation of the localized adhesion onto epithelial cells. In this study, we demonstrate that clinical isolates of serotypes O128ab:H2 and O119:H2 contain a ca. 13-kb deletion in the bfp operon, resulting in a lack of expression of these pili. An IS sequence with homology to the IS66 of Agrobacterium tumefaciens replaced the deleted bfp genes. These results suggest that the … Show more

“…These strains were negative for EAF and bfpA probes, but they were positive for perA , an EAF gene [21]. Therefore, like some other atypical strains that have been described in the literature [28-30], these strains carry vestiges of the EAF plasmid.…”

“…It is important to point out that EPEC strains of serotypes O128:H2 and O119:H2 contain a pEAF with defective bfp operons, which contain part of the bfpA gene but have the rest of the bfp gene cluster deleted. Thus, they are classified as aEPEC 12, 75. Most aEPEC produce adherence patterns categorized as LA-like, with loosened microcolonies compared to those of the tEPEC LA pattern 12, 76, 77.…”

“…For example, serotypes O119:H2 and O128:H2 react with the bfpA probe but do not have a true EAF plasmid. These serotypes have a 100-MDa plasmid that does not contain the bfp operon and consequently does not produce BFP ( 19 ). In contrast, some O142:H6 strains do not react with the EAF probe but produce BFP and show a typical localized adherence (LA) pattern.…”
