Lactobacillus acidophilus K301 Inhibits Atherogenesis via Induction of 24 (S), 25-Epoxycholesterol-Mediated ABCA1 and ABCG1 Production and Cholesterol Efflux in Macrophages

Abstract: Lactobacillus acidophilus species are well-known probiotics with the beneficial activity of regulating cholesterol levels. In this study, we showed that L. acidophilus K301 reduced the level of cholesterol through reverse transport in macrophages. L. acidophilus K301 upregulated the mRNA and protein levels of genes such as ATP-binding cassette A1 (ABCA1) and ATP-binding cassette G1 (ABCG1) under the control of liver X receptor (LXR), resulting in increased apoA-I-dependent cholesterol efflux in phorbol 12-myri… Show more

“…TG has now been recognized as a strong marker for CVD and diabetes, as well as NAFLD [4]. Probiotics have been reported to exert their lipid-modulating effect by regulating the host genes responsible for lipid metabolism [20,24,25]. In this study, it was observed that L. plantarum DR7 and L. fermentum DR9 significantly downregulated the gene expression of SCD1 in the liver, where the former showed a more prominent effect.…”

“…The differentiation of THP-1 to macrophages was confirmed by increasing CD36 and decreasing CCR2 (Fig. 1C) [14]. In addition, cell viability was not affected by the ligands used in this study (Fig.…”

“…For example, meta-analysis of 15 studies with 788 subjects showed effective lowering of cardiovascular risk factors such as total cholesterol, LDL-C and inflammatory markers [ 107 ], conclusions that are supported by another similar meta-analysis [ 108 ]. Studies in mouse model systems have also shown attenuation of atherosclerosis by probiotic bacteria via multiple mechanisms, including induction of tolerogenic dendritic cells [ 109 ], stimulation of cholesterol efflux [ 110 ] and reduction in both intestinal cholesterol uptake and vascular inflammation [ 111 , 112 ].…”
