Large-Scale Expansion of Porcine Adipose-Derived Stem Cells Based on Microcarriers System for Cultured Meat Production

Abstract: Cultured meat is an innovative meat-production technology that does not rely on animal husbandry. As a new food component, cultured fat is of great significance to cultured meat. In this study, we isolated adipose-derived stem cells (ADSCs) and identified the purity by immunofluorescence staining of ADSC-specific surface marker proteins CD44 and CD29 and showed that most of the cells were positive for CD29 and CD44. In addition, we detected the expression of FABP4 and Plin1 to confirm that ADSCs differentiated… Show more

“…Previous studies have shown that many primary preadipocytes can grow on Cytodex 1 microcarriers 18,20 . However, the process of digesting cells from microcarriers is required since Cytodex1 is inedible, increasing workloads and costs.…”

“…For large-scale expansion of seed cells for cell-cultured meat, acclimating to 3D-scale culture is critical. Several primary preadipocyte cell lines have been reported to be amplified on microcarriers in suspension bioreactors to increase scalability 18,20 . We reported that ISP-4 could achieve large-scale expansion on microcarriers at the density of 18 × 10 5 cells per ml.…”

“…Therefore, further work should focus on adapting Pig#4 to anchorage-independent growth. Moreover, the primary preadipocytes need to be harvested from microcarriers before adipogenic differentiation 18,20 . Although it remains unclear whether it is because the microcarriers are inedible, or these primary cells cannot be differentiated on the microcarriers, the inadequacy of these protocols is apparent, which increases the labor costs.…”

“…Many cell sources have been explored, including muscle satellite cells, adipose-derived stem cells, multipotent mesenchymal stem cells, induced pluripotent stem cells, transgenics, and immortalized cells [2]. Among them, multipotent mesenchymal stem cells (stem cells) stand out as promising candidates due to their abilities of self-renewal, differentiation, and their relatively easy acquirement from multiple types of tissues [1,11]. Stem cells or progenitor cells used in cultivated meat in current literature are mostly isolated from tissue biopsies via enzymatic digestion.…”
