Paclitaxel and baccatin III-producing cells of Taxus baccata were immobilized within Ca(2+)-alginate beads. Under established optimum conditions for the biosynthesis of both taxanes, the yields of paclitaxel and baccatin III in shake-flask cultures of free cells increased by factors of up to 3 and 2, respectively, in the corresponding cultures of immobilized cells. Although the scale-up from shake-flask to bioreactor culture usually results in reduced productivities when both free and immobilized cells were grown in the same optimum conditions in three different bioreactor types (Stirred, Airlift, and Wave) running for 24 days in a batch mode and with the system optimized in each case, there was a considerable increase in the yields of paclitaxel and baccatin III. Among the reactors, the Stirred bioreactor was the most efficient in promoting immobilized cell production of paclitaxel, giving a content of 43.43 mg.L(-1) at 16 days of culture, equivalent to a rate of 2.71 mg.L(-1).day(-1). To our knowledge, the paclitaxel productivity obtained in this study is one of the highest reported so far by academic laboratories for Taxus species cultures in bioreactors.

A simple and accurate RP-HPLC method with pentafluorophenyl (PFP) column was developed for the simultaneous determination of six taxoids, i.e. paclitaxel, 10-deacetylbaccatin III (10-DAB III), 7-xylosyl-10-deacetyltaxol (7-xyl-10-DAT), 10-deacetyltaxol (10-DAT), cephalomannine and 7-epi-10-deacetyltaxol (7-epi-10-DAT), in the extracts from the needles of three Taxus species. The mobile phase consisted of acetonitrile (A) and water (B), and the extracts were separated using gradient elution program: 30% A at the first 7 min, and then ramped to 42% A at 8 min, held until 38 min. The developed method was validated with satisfactory precision (RSD < 2.61%), repeatability (RSD < 2.92%) and recovery (95.19-104.47%). The above taxoids in the extracts of Taxus cuspidata, T. chinensis and T. media were analyzed with the developed RP-HPLC method, and the results showed that the contents of different taxoids in three mentioned species were distinct. Maximal amounts of 10-DAB III, 7-xyl-10-DAT and 7-epi-10-DAT appeared in T. chinensis, while T. media possessed the highest content of 10-DAT, cephalomannine and paclitaxel. The developed method is accurate and efficient. It can be reliably used in the improved determination of taxoids for the quality control of Taxus species.

RESUMENEl árbol del tejo (Taxus spp.) es una gimnosperma dicotiledónea que pertenece a la familia Taxaceae; se distribuye en America, Europa y Asia. Taxus spp. contiene en la corteza y en las hojas, alcaloides diterpénicos conocidos como taxoides. Éstos se caracterizan químicamente por la presencia de un esqueleto de taxano y un anillo de oxetano, tal como el taxol, que es utilizado en el tratamiento del cáncer de ovario, seno, próstata y pulmón; enfermedad que actualmente es una de las principales causas de muerte a nivel mundial. El contenido de taxol es bajo en los árboles, por tanto, es necesario el uso de alternativas biotecnológicas como lo es el cultivo in vitro de tejidos vegetales para incrementar la cantidad de taxoides y preservar el recurso natural. En esta revisión científica se reportan las características químicas de los taxoides, el mecanismo de acción biológico y semisíntesis orgánica del taxol, se detalla también la ruta biosintética de los principales taxoides, así como las principales enzimas involucradas para la obtención de éstos a nivel bioquímico y, finalmente, se indica de manera general la producción de taxoides en cultivos in vitro. ABSTRACTThe yew tree (Taxus spp.) is a dicot gymnosperm belonging to the Taxaceae family and it can be found in America, Europe and Asia. These trees have in their bark and leaves diterpenic alkaloids known as taxoids, which are chemically characterized by the presence of a taxane skeleton and an oxetane ring such as taxol, which is used in the treatment of ovarian, breast, prostate and lung cancer; disease that is currently one of the leading causes of death worldwide. Taxol content is low in trees, so it is necessary to use biotechnology alternatives such as in vitro cultivation of plant tissue to increase the amount of taxoids and preserve the natural resource. The chemical characteristics of taxoids, the mechanism of the biological action and the semi-synthetic organic of taxol were reported in this scientific review; as well as, the biosynthetic pathway of major taxoids and the main enzymes involved in obtaining taxoids at biochemical level and, finally the production of taxoids in vitro cultures is indicated.
