Large-scale identification of differentially expressed genes during neurogenesis

Abstract: We report here a modified mRNA differential display method and its application for the analysis of differential gene expression in NGF-treated PC12 cells and in embryonic rat spinal cord. The optimized protocol is based on low fidelity priming of multiple cDNAs followed by high fidelity amplification. In PC12 cells induction by nerve growth factor (NGF) altered the expression of 4% of the 466 transcripts evaluated. During neurogenesis of the spinal cord we found that 30% of the 288 examined products changed. T… Show more

“…Using a number of variations in culture conditions and/or genetic modifi cations, cultured neuronal stem cells can differentiate into different neuronal progeny. The complex events leading to the generation of different phenotypes via distinct developmental programs can be studied systematically with transgenic technology and microarray techniques [4][5][6]8] . However, ultimately the regulatory functions are carried out by the cellular protein constituents, and proteomic approaches are most appropriate to determine protein content, stability and subcellular localization, posttranslational modifications and protein-protein interactions.…”

“…Differentiation and maturation of stem cells comprises complex serial events, which guide the undifferentiated cells to different lineages via distinctive developmental programs (Chung et al, 2005;Hoffrogge et al, 2006;Kwak et al, 2006). Such regulatory changes can be studied systematically with transgenic technology and microarrays (Pazman et al, 2000;Böttcher et al, 2003;Gurok et al, 2004;Pahnke et al, 2004). The process of differentiation entails changes in types and amount of proteins expressed by the stem cell.…”
