Large-scale Micropropagation of Aloe vera

Abstract: A protocol for large-scale Aloe vera (L.) Burm. f. production was established using micropropagation of apical buds. The effects of two chlorine-based disinfectants were evaluated on the survival of the explants in different treatments in a semisolidified Murashige and Skoog (MS) medium in the presence of 6-benzylaminopurine (6-BAP; 2 mg·L−1). During 120 days, 136 green apical shoots bearing axillary buds were multiplied four times at 30-day intervals in … Show more

“…Explants were sterilized sequentially by washing with running tap water for 30 minutes; dipping and stirring in a mixture of ridomile, kocide, and baylaton (1 : 1:1 v/v) for 10 minutes; dipping and stirring in Tween 20 solution for 10 minutes; rinsing with tap water four times; rinsing with distilled water two times to remove traces of detergent; dipping in 70% ethanol for 30 seconds; dipping and stirring in 20% sodium hypochlorite for 10 minutes; and rinsing with distilled water three times. en, they were further sterilized in a laminar air flow cabinet by surface sterilization using freshly prepared HgCl 2 (0.25% w/ v aqueous solution) for 5 minutes followed by thorough washing using sterile distilled water to remove traces of HgCl 2 [15][16][17][18][19].…”

“…This success has been positively correlated to the dimensions of the ex vivo microplants: medium (6–8 cm long bearing 4–5 leaves) and large (13–16 cm long bearing 5–7 leaves) sizes have been found to be significantly different from small (3–4 cm long bearing 2–3 leaves) microplants (de Oliveira et al, 2009). …”

“…Various types of explants are used to get axillary branching: meristem in order to obtain virus-free aloe (Campestrini et al, 2006; Das et al, 2010; Rizwan et al, 2014), shoot tips (Meyer and Van Staden, 1991; Aggarwal and Barna, 2004; Ahmed et al, 2007; Supe, 2007; Hashemabadi and Kaviani, 2008; Zakia et al, 2013; Amoo et al, 2014; Molsaghi et al, 2014), axillary shoot segments (Singh et al, 2009; Shekhawat et al, 2013), shoot bud (Kanwar et al, 2015), apical buds (de Oliveira et al, 2009; Gupta et al, 2014), buds from underground stems (Liao et al, 2004), stem nodes (Singh and Sood, 2009), rhizomatous stems (Gantait et al, 2010, 2011), and suckers (Nayanakantha et al, 2010b). …”
