Laser Microdissection

Abstract: Laser microdissection (LM) offers a relatively rapid and precise method of isolating and removing specified cells from complex tissues for subsequent analysis of their RNA, DNA, protein or metabolite content, thereby allowing assessment of the role of different cell types in the normal physiological or disease processes being studied. In this unit, protocols for the preparation of mammalian frozen tissues, fixed tissues, and cytologic specimens for LM, including tissue freezing, tissue processing and paraffin … Show more

“…The POL membrane slides with collected sections were dehydrated in a desiccation chamber with Drierite (with indicator) for 30–45 min as described (Morrison, Bailey, & Kulesa, 2012; Morrison, Box, McKinney, McLennan, & Kulesa, 2015). Laser microdissection (Frost, Eltoum, Siegal, Emmert‐Buck, & Tangrea, 2015) was performed using a Leica LMD 600 in the Cambridge Advanced Imaging Centre (University of Cambridge, Cambridge, UK).…”

“…Laser microdissection is a technique that utilizes a focused beam to isolate cell populations or individual cells from tissue sections under microscopic guidance [ 1 , 2 ]. Prior to laser microdissection, tissues need to be embedded into blocks before they are sectioned by microtome.…”

“…And several different collection methods have been developed combined with laser microdissection. Contact collection methods require customized tube caps, including the silicone-filled adhesive cap and the transfer cap covered with a layer of thermoplastic film [ 15 ]. Compared with contact collection, non-contact collection has greater potential to avoid pollution.…”
