Lateral flow immunoassay strips based on europium(III) chelate microparticle for the rapid and sensitive detection of Trichinella spiralis infection in whole blood samples of pigs

Abstract: Trichinellosis is a major food-borne parasitosis caused by ingesting raw or semi-raw meat products from pigs infected with Trichinella spiralis (T. spiralis). Although China is the largest consumer of pork in the world, the current diagnostic method of T. spiralis is exclusively performed in a laboratory setting, due to its complexity and laborious procedure. Here, in order to solve the detection problems in the pig breeding industry, a rapid, sensitive, and on-site serological diagnosis method was developed. … Show more

“…The mixture was again centrifuged to remove the supernatant and the pellet was added with fluorescent probes dissolved in a preserved buffer (25 mM/L Tris containing 0.05% Tween-20, 0.15 mM/L NaCl, 0.05% Pro Clin 300, 1% BSA, and 5% Trehalose). Similar procedures were followed for the microspheres conjugation with goat anti-rabbit IgG [ 25 ].…”

“…Firstly, the sample gasket materials (SB08, 8964, XQY8, ST17, SB06, RB45, 6613, and RB65) and made of glass fiber and polyester fiber were evaluated for their applicability in ICS. Secondly, TSOL18 and fluorescent microspheres coupled at 1:5, 1:10, 1:15, and 1:20 were evaluated [ 25 ].…”

“…The conventional immunochromatographic assay is convenient to detect pathogen infection within a few minutes, but the sensitivity needs to be improved [17]. Fortunately, with the application of nanomaterials, the detection signal of the test strip can be strongly enhanced, and the detection sensitivity can be greatly improved [18][19]. Eu (III) nanoparticles have the characteristics of a large Stokes shift (>150 nm), long fluorescence lifetime and low interference from background fluorescence, thereby leading to improved sensitivity [20][21][22].…”

“…To improve the implementation of LFAs, new fluorescent labels have been developed to increase the overall sensitivity. Among them, lanthanide chelate-doped polystyrene nanoparticles have gained attention due to their higher fluorescence than free dye molecules, low variation, and long-lifetime fluorescence, also providing the advantages of common colored polystyrene nanoparticles [ 21 , 22 , 23 , 24 ].…”
