Layer-by-Layer Investigation of Ultrastructures and Biomechanics of Human Cornea

Abstract: The cornea is an avascular, innervated, and transparent tissue composed of five layers: the epithelium, Bowman’s layer, stroma, Descemet’s membrane, and endothelium. It is located in the outermost fraction of the eyeball and is responsible for the refraction of two-thirds of light and protection from external mechanical damage. Although several studies have been done on the cornea on the macroscopic scale, there is a lack of studies on the micro-nanoscopic scale, especially an analysis evaluating the cornea la… Show more

“…Tissue shape and mechanics, for example, are critical for regulating organ function. AFM can capture high-resolution images and connect them with mechanical and viscoelastic properties, making it an ideal tool for studying blood cells, cancer cells, , and viral particles, among other biological systems . It is worth noting that AFM can be utilized in the physiological circumstances of cells or biomolecules.…”

“…During the separation process, the zero deflection of the cantilever beam also does not mean that the probe is separated from the sample. Due to the adhesive forces between the probe tip and the sample, the cantilever beam deforms and forces in the opposite direction before the two are truly separated (Dias Rates et al, 2022). The “instantaneous” separation occurs when the probe continues to move away from the sample and overcomes the adhesive forces (Holuigue et al, 2022).…”

“…The lucidity of the cornea is due to the complicated balance between the layers and the cellular components of the cornea. 1 Cornea is an immune-privileged structure, which provides protection to the eye and is the reason for successful corneal transplants. 1,2 Corneal neovascularization occurs in various pathologies and can result in compromise of corneal transparency.…”
