Lectin histochemistry of the mast cell: Heterogeneity of rodent and human mast cell populations

Abstract: Mast cell granules contain a variety of N-linked saccharides. Heterogeneity of the expression of these saccharides in mast cells was studied in rodent and human tissues which were so selected as to contain all the mast cell subsets previously identified using other criteria. Dermal and intestinal mucosal mast cells were stained with lectins using an avidin-biotin system. It was found that dermal and subepidermal mast cells in the rat and mouse, and mucosal and dermal human mast cells showed very similar lectin… Show more

“…The first segment was fixed in Carnoy's solution, cleared in xylene, and embedded in paraffin blocks. Transverse sections (5 ìm) were cut and stained with Alcian Blue-Safranin O according to Roberts, Jones & Stoddart (1990). Mast cell evaluation in mucosa and sub-mucosa was expressed in number per high-powered field (HPF, using 20 ² objective).…”

“…4 The affinity of lectins for carbohydrates has been applied to the characterization of mast cell populations in humans, rodents, and dogs. 2,5,14,17,32 Although special stains have been used to diagnose canine mast cell tumors, 1,12,24,31 a systematic comparison has not been done. The purposes of this study Received for publication October 12, 1993. were to evaluate the use of histochemical, affinity, and immunohistochemical stains in formalin-fixed tissues in diagnosing mast cell tumors, especially poorly differentiated tumors, and to determine if the stains would differentiate them from histiocytomas.…”

“…There is moderately strong membranous, goblet cell and stromal staining with e-PHA in this section from distal colon (e-PHA x 2100). other normal organs and pathological processes (Jones & Stoddart, 1986;McMahon et al, 1989;Roberts et a]., 1990). Other factors, including type and duration of tissue fixation and tissue processing, may also be important in the analysis of such variations.…”
