Lentivirus-mediated RNAi silencing targeting ABCC2 increasing the sensitivity of a human nasopharyngeal carcinoma cell line against cisplatin

Abstract: Background: High resistance to drug is taken as a characteristic of human tumors, which is usually mediated by multidrug resistance-associated genes. ABCC2, an ATP-binding cassette multidrug resistance transporter, is found to be expressed in a variety of human cancers. In this study the effect of a RNAi construct targeting ABCC2 on the chemosensitivity of NPC cell line CNE2 against cisplatin was investigated.

“…In clinical practice, NPC is treated by radiation, but the therapeutic effect is not satisfactory (8)(9)(10). In recent years, many investigations have focused on extracting active ingredients from natural plants to prevent and treat cancer and to investigate their anti-cancer mechanism (11,12).…”

“…cDNA was synthesized using RT reagents (Takara, Dalian, China) from 1 mg of total RNA using oligo(dT) 15 in 20 ml reaction volume. Real-time PCR was used to measure the expression of MMP2 mRNA using SYBR Pre-mix Ex Taq according to the manufacturer's protocol (Takara) with an Mx3000P real-time PCR system (Stratagene, La Jolla, USA) as described previously [18]. The sequence for sense primer was 5 0 -ACCAGCTGGCCTAGTGATGATG-3 0 , and for anti-sense primer was 5 0 -GGCTTCCGCATGGTCTCG ATG-3 0 .…”

“…The combination of radiotherapy and adjuvant chemotherapy is the prevalent treatment of NPC, and the most chemotherapeutic agents used for treating NPC are cisplatin based [18]. Nevertheless, the acute and late toxicities, as well as resistance of the chemotherapies have significantly inscreased [19,20]. Therefore, there is a need for the development of less toxic and more effective agents for NPC treatment.…”
