Leptin is an effective treatment for hypothalamic amenorrhea

Abstract: Hypothalamic amenorrhea (HA) is associated with dysfunction of the hypothalamic-pituitary-peripheral endocrine axes, leading to infertility and bone loss, and usually is caused by chronic energy deficiency secondary to strenuous exercise and/or decreased food intake. Energy deficiency also leads to hypoleptinemia, which has been proposed, on the basis of observational studies as well as an open-label study, to mediate the neuroendocrine abnormalities associated with this condition. To prove definitively a caus… Show more

“…Therefore, it would be reasonable to assume that leptin may be applicable for clinical use in other non--lipodystrophy leptin-deficient conditions. Indeed, leptin has previously been investigated in hypothalamic amenorrhea with positive results (40,41). Finally, age-and sex-specific effects cannot be discounted as possible sources of variation and error, and future clinical trials should investigate these aspects, as some studies assessed here included only children, and others only women ( Table 2).…”

“…In three females an ovulatory cycle occurred; others also showed evidence for an improved reproductive function [13]. A subsequent randomized, double-blinded placebocontrolled trail of 36 weeks demonstrated similar beneficial effects [14].…”

“…One of the results of these processes is central amenorrhea. Administration of leptin in these patients results in recovery of menstruation 28 . Intravenous administration of leptin in human dietary intake (weight reduction programs) resulted in weight reduction compared to placebo 29 .…”
