Leptosphaeria maculans, a fungal pathogen of Brassica napus, secretes a subtilisin-like serine protease

Abstract: Leptosphaeria maculans, a fungal pathogen of Brassica napus, secretes large amounts of a 28 kDa protein (SP2) in liquid culture. This protein shows high sequence similarity to secreted serine proteases from other ascomycetes and is the major component of culture filtrate with protease activity, as analysed on casein zymogels. The sp2 gene is expressed during infection of B. napus cotyledons when L. maculans hyphae are growing between mesophyll cells, as well as at later stages when the fungus invades the vascu… Show more

“…This research, along with studies on particular genes or randomly mutated genes Idnurm and Howlett, 2003;Idnurm et al, 2003b;Meyer et al, 2004) has resulted in additional annotated genes published on international databases e.g. opsin, isocitrate lyase, cyanide hydratase, neutral trehalase, secreted proteins sp1 and sp2, G a subunit Idnurm and Howlett, 2002;Wilson et al, 2002;Idnurm et al, 2003b;Wilson and Howlett, 2005; M. “…Sexton et al (2000) have cloned a gene encoding endo-PG, pg1, and two genes encoding cellulases, cel1 and cel2, in L. maculans. The sp1 and sp2 genes are also expressed in L. maculans during the infection of B. napus plants, and later the gene secretes a serine protease with protease activity (Wilson and Howlett 2005). The Lmpma 1 gene of L. maculans encodes a plasma membrane H+-ATPase isoform, which is essential for pathogenicity toward oilseed rape (Remy et al 2008).…”

“…Thirteen of the proteins found were hypothetical proteins with unknown functions. Among the detected proteins the previously described secreted proteins 1 29 and 2 30 were also found. They occurred among the 18 proteins that were detected in all three protein fractions pF1-pF3 after IEF ( Figure 10A).…”

“…They are also highly expressed during infection in B. napus . While the function of sp1 is not clear, sp2 has been identified as a subtilisin‐like serine protease and proposed as having firstly a nutritional role by degrading proteins in the plant apoplast during the intercellular growth of L. maculans , and secondly during the later stages, when L. maculans kills plant cells, sp2 may contribute to the degradation of plant cell walls . However, to our knowledge no report about their elicitation activity exists.…”
