Leucine 42 in the Fibronectin Motif of Streptokinase Plays a Critical Role in Fibrin-independent Plasminogen Activation

Abstract: The NH 2 terminus (residues 1-59) of streptokinase (SK) is a molecular switch that permits fibrin-independent plasminogen activation. Targeted mutations were made in recombinant (r) SK1-59 to identify structural interactions required for this process. Mutagenesis established the functional roles of Phe-37and Glu-39, which were projected to interact with microplasmin in the activator complex. Mutation of Leu-42 (rSK1-59 L42A ), a conserved residue in the SK fibronectin motif that lacks interactions with micropl… Show more

“…7), which binds and activates Plg, thereby facilitating invasion of bacteria through the host tissue barriers. The stretch around Val 19 in the N terminus of streptokinase is implicated in Plg binding (35,37), and that around Leu 42 is involved in activation of Plg (38). From the corresponding sequences within the N terminus of CD222, we designed two peptides, termed pepB and pepC, respectively.…”

“…Two peptides were synthesized: pepB, derived from the stretch similar to the site in streptokinase playing a crucial role for Plg binding (37); and pepC, derived from the sequence corresponding to the site of streptokinase involved in Plg activation rather than binding (38) (Fig. 7).…”

“…Consequently, the cDNA for the SK␤swap chimera was generated in such a way that the codons for residues 146 -281 of SK were replaced by the codons for residues 126 -281 of SUPA by PCR with an overlap extension as we have described previously (33). The SK␤swap chimera cDNA was ligated into the pMALc vector (New England Biolabs, Beverly, MA) for expression as a maltose-binding fusion protein in bacteria (20).…”

“…Wild type SK, a deletion SK mutant (SK⌬59) lacking the first 59 amino acids, SK1-59, and SK1-59(L42A) were cloned as described (9,11,22). Tryptophan 6 was mutated to alanine (W6A) in the SK1-59 and SK1-59(L42A) by PCR to reduce intrinsic fluorescence.…”
