(-)-Leucophyllone, a Tirucallane Triterpenoid from Cornus walteri, Enhances Insulin Secretion in INS-1 Cells

Abstract: Phytochemical examination of the MeOH extract from the stems and stem bark of Cornus walteri (Cornaceae) led to the isolation and verification of a tirucallane triterpenoid, (-)-leucophyllone, as a major component. Its structure was elucidated using NMR spectroscopy and liquid chromatography–mass spectrometry. The effect of (-)-leucophyllone on insulin secretion in INS-1 cells was investigated. (-)-Leucophyllone increased glucose-stimulated insulin secretion (GSIS) at concentrations showing no cytotoxic effect… Show more

“…Insulin secretion decreased in oxidative stress-induced pancreatic β cells, but recovered under co-treatment with EA ( Figures 4A, B ). Flavonoids upregulate the protein expression (including PDX-1 expression) involved in pancreatic β cell function ( Lee et al, 2021 ). Protein expression of PDX-1 increased markedly following EA treatment ( Figures 4C, D ).…”

“…Shionone is one of the important triterpenoid components of phytochemicals present in AT, which is considered a content determination index for RAS quality and is believed to be primarily responsible for its pharmacological properties. Triterpenoids are important phytochemicals known to possess a variety of important medicinal properties that can make them useful as therapeutic agents [ 2 , 3 ].…”
