Levels of bovine papillomavirus RNA and protein expression correlate with variations in the tumorigenic phenotype of hamster cells

Abstract: Three independent cell lines were established from primary cultures of LSH hamster embryo cells infected with bovine papillomavirus type 1 (BPV-1). Although these cell lines differed in their in vitro saturation densities, none was capable of colony formation in soft agar. Interestingly, two cell lines (BPV-HE1 and BPV-HE3) were tumorigenic in nude mice, syngeneic hamsters, and allogeneic hamsters, whereas BPV-HE2 was not. All three cell lines contained similar numbers of the BPV-1 genome (approximately 50 to … Show more

“…here imply that the phenotype of cultured cells stably transformed by BPV-1 is not an all-or-nothing phenomenon but rather is correlated with the dose of viral gene products. This conclusion was previously suggested by the behavior of BPV-1-transformed hamster cells and C127 cells harboring a low-copy-number BPV-1 mutant (4,39). However, a recent study of BPV-transformed C127 cell clones failed to reveal such a correlation (37).…”

“…BPHE-1 cells were obtained from A. Lewis (National Institutes of Health, Bethesda, Md.) (42). C127 clone C cells were obtained from W. Vass (National Institutes of Health), and BHK-21 cells were from the American Type Culture Collection.…”

“…Subnuclear localization of BPV capsid proteins. BPHE-1 is a hamster fibroblast cell line that is latently infected with multiple copies of autonomously replicating BPV genomes and expresses the nonstructural viral proteins (57). We used the SFV expression system to introduce the L2 minor capsid protein into BPHE-1 cells and localized the L2 protein by immunofluorescence staining and laser scanning confocal microscopy.…”

“…Cell lines. BPHE-1 cells, obtained from Andrew Lewis Jr. (NIH), were grown in Dulbecco's modified Eagle's medium supplemented with antibiotics and 10% fetal calf serum (57). BHK-21 cells were grown in Glascow's medium supplemented with 10% tryptose phosphate broth, antibiotics, nonessential amino acids, HEPES, and 5% fetal calf serum.…”
