Summary The enzymatic activities of aminotransferases AST and ALT have been measured in the semen of a group of 98 subjects undergoing fertility studies, correlated with spermatic density and motility. An AST/ALT ratio was obtained for all patients. The results indicate that there exists a correlation between both enzymes' activities and spermatic density and motility. In two cases of excretory azoospermia, an elevated AST/ALT relation was obtained, while in two cases of secretory azoospermia a low AST/ALT ratio was seen. Aminotransferase values can thus be considered a useful piece of data in the evaluation of seminal quality, and the AST/ALT ratio used more specifically in the discrimination of the various types of azoospermia. Die Aktivität der Aspartat‐Amino‐Transferase und Alanin‐Amino‐Transferase im menschlichen Sperma bei männlichen Fertilitätsuntersuchungen Zusammenfassung Die Enzymaktivitäten der Aminotransferasen AST und ALT, die im Sperma von 98 Personen gemessen wurden, die sich einer Fertilitätsuntersuchung unterzogen, korrelierten mit der Spermatozoendichte und der Beweglichkeit. Eine Korrelation AST/ALT fand sich bei allen Patienten. In zwei Fällen mit exkretorischer Azoospermie fand sich eine hohe AST/ALT‐Relation, während in zwei Fällen mit sekretorischer Azoospermie eine niedrige AST/ALT‐Relation nachweisbar war. Die Aminotransferasewerte stellen somit ein wertvolles Teilergebnis bei der Bewertung der Spermaqualität dar, und die AST/ALT‐Korrelation erlaubt eine spezifischere Unterscheidung der verschiedenen Typen von Azoospermie.

Determinations of aspartate and alanine aminotransferase activity in human semen were carried out by the spectrophotometric and colorimetric methods. It was found that the occurrence of interfering substances in semen makes the spectrophotometric method unsuitable without prior partial purification of the enzyme. The mean activity determined colorimetrically in 417 samples of human semen was 320 Sigma-Frankel units/ml (s.e.m. \m=+-\8,range 10 to 1070) for aspartate aminotransferase and 30 units/ml (s.e.m. \ m=+-\ 3, range 0 to 97, n = 40) for alanine aminotransferase. As regards aspartate aminotransferase, the activity of this enzyme was highest at +60\s=deg\ C. Pyridoxal phosphate increased the activity, particularly if added to semen samples with initially low activity. Gel electrophoresis of seminal plasma showed one band migrating towards the anode. However, in homogenized spermatozoa the bulk of enzyme migrated towards the cathode and only a weak band could be demonstrated on the side of the anode.

Determinations were made of glutamic-oxaloacetic transaminase activity in 135 ejaculates, ten testicular biopsy specimens, and post-mortem material consisting of testes, epididymides, seminal ducts, prostate glands and seminal vesicles. Transaminase levels in semen appeared to be directly related to sperm concentration. Lowest values were observed in azoospermic and aspermic semen. The various organs which compose the male reproductive tract did not differ much in transaminase activity.
