Light and Fungal Elicitor Induce 3-Deoxy-d-arabino-Heptulosonate 7-Phosphate Synthase mRNA in Suspension Cultured Cells of Parsley (Petroselinum crispum L.)

Abstract: Light and fungal elicitor induce mRNA encoding 3-deoxy-Darabino-heptulosonate 7-phosphate (DAHP) synthase in suspension cultured cells of parsley (Petroselinum crispum L.). The kinetics and dose response of mRNA accumulation were similar for DAHP synthase and phenylalanine ammonia-lyase (PAL). Six micrograms of elicitor from Phytophthora megasperma f. glycinia gave a detectable induction within 1 hour. Induction of DAHP synthase and PAL mRNAs by light was transient, reaching maximal levels at 4 hours and retum… Show more

“…Vol. 108, 1995 Conn, 1989;Henstrand et al, 1992). The induction of DAHP synthase gene expression in response to severa1 of these environmental stimuli parallels a similar induction of PAL mRNA (Dyer et al, 1989;Henstrand et al, 1992), suggesting that the synthesis of primary aromatic amino acids and secondary aromatic compounds might be regulated in concert at the transcriptional level.…”

“…DAHP synthase is transcriptionally induced in response to a number of environmental stresses (Pinto et al, 1988;Dyer et al, 1989;Keith et al, 1991;Henstrand et al, 1992). Elicitor and light increase DAHP synthase activity in parsley (McCue and Conn, 1989), and wounding increases DAHP synthase activity, polypeptide, and mRNA in potato (Dyer et al, 1989;Muday and Herrmann, 1992).…”

“…This may include increased availability of precursors and cofactors, including erythrose-4-phosphate, phosphoenolpyruvate, NADH, and ATP, which are produced in chloroplasts from photosynthesis. As a likely consequence of this, the shikimate pathway has been reported to be light regulated in some plants (19,53). Furthermore, Euglena gracilis has been reported to have a cytosolic pathway used when grown in darkness but a plastidic pathway used when grown in light (36).…”

“…The localisation to chloroplasts make light and photosynthesis plausible candidates for regulation of the pathway, and in plants light may be the main regulator of the DAHPS. Light has been shown to upregulate DAHPS at the transcriptional level in parsley cell cultures (Henstrand 1992). However, although the shikimate pathway is located in the plastids, non-green tissue, like roots and flowers, contain the greatest amount of mRNA for the inducible isoenzymes of the pathway (Weaver and Herrmann 1997).…”
