“…Kim and Hong reported about an enhancement of affinity of a humanized monoclonal antibody after light chain shuffling (Kim and Hong 2007). For allergen-specific human IgE antibodies there is some evidence that the heavy chain determines allergen-specificity (Laffer et al 2001;Rojas et al 2004;Christensen et al 2009;Gadermaier et al 2010b). It is therefore quite possible, that the affinity of allergen-specific antibodies can be increased by shuffling of light chains.…”

“…To probe if hypoxia also triggers increased expression of NeuGc GM3 in HeLa cells, flow cytometry experiments were performed. For detection, we used the murine antibody 14F7, which specifically recognizes NeuGc GM3 and can discriminate it from the very similar NeuAc derivative [20,[37][38][39]. This antibody was earlier subjected to glycoarray screening by the Functional Glycomics Consortium (www.functionalglycomics.org), with negative results, even for the NeuGc GM3 trisaccharide (data not shown), indicating that it is indeed very specific and furthermore requires the ganglioside's sphingolipid part for antigen recognition.…”

“…25,31 Soluble antibody fragments from the selected clones were usually produced by induction with IPTG in the absence of helper phage. 32 In the present study, the supernatant of lysate was used for the identification of the positive clones.…”
