Light-independent regulation of algal photoprotection by CO2 availability

Abstract: Photosynthetic algae have evolved mechanisms to cope with suboptimal light and CO2 conditions. When light energy exceeds CO2 fixation capacity, Chlamydomonas reinhardtii activates photoprotection, mediated by LHCSR1/3 and PSBS, and the CO2 Concentrating Mechanism (CCM). How light and CO2 signals converge to regulate these processes remains unclear. Here, we show that excess light activates photoprotection- and CCM-related genes by altering intracellular CO2 concentrations and that depletion of CO2 drives these… Show more

“…ROC75 and the CCM transporters LCIA and HLA3 are separated from the CCM cluster (Figure 1B). Taken together, the overall expression patterns in unbiased large-scale data reveal a diverse behaviour of the PP and CCM genes known to be co-expressed in some conditions (Ruiz-Sola et al, 2023;Arend et al, 2023). Co-expression with known TFs involved in these processes is limited, which indicates post-transcriptional modification, and/or non-linear, and/or combinatorial interactions.…”

“…Since TFs known to be involved in PP typically also mount a transcriptional phenotype for genes involved in the CCM (Ruiz-Sola et al, 2023;Arend et al, 2023), we tested the candidate TFs for changes in CCM gene expression (Figure 3) in response to low CO2 supply (Figure 3A, red line).…”

“…Ionomic analysis was performed directly on snow water acidified with 0.65% (w/v) nitric acid and on snow water that has been concentrated 10 to 12.5 fold (freeze-drying followed by solubilization in 0.65% (w/v) nitric acid). Samples were analyzed using an iCAP RQ quadrupole mass instrument (Thermo Fisher Scientific GmbH, Germany) operating in the standard mode (for 11 B, 23 Na, 24 Mg, 25 Mg, 27 Al, 31 P, 39 K, 206 Pb, 208 Pb) and the collision mode with helium as a cell gas (for 31 P, 39 K, 44 Ca, 52 Cr, 53 Cr, 55 Mn, 56 Fe, 57 Fe, 59 Co, 63 Cu, 65 Cu, 64 Zn, 66 Zn, 69 Ga). Quantification was done using standard curves, corrected using the internal standards 103 Rh and 172 Yb, and molar concentrations were calculated using snow liquid water content of 15 mass%.…”

“…See supplementary Methods.Fluorescence-based measurements and statistical analysis. Fluorescence-based photosynthetic parameters were measured with a pulse modulated amplitude fluorimeter (MAXI-IMAGING-PAM, HeinzWaltz GmbH, Germany) as described in66 . Prior to the onset of the measurements, cells were acclimated to darkness for 15 min.…”
