Light-regulated betacyanin accumulation in euhalophyte Suaeda salsa calli

Abstract: Suaeda salsa calli cultured in darkness for 28 days were transferred to Murashige and Skoog (MS) media containing 0.5 mg l -1 Thidiazuron (TDZ) and 1.0 mg l -1 naphthaleneacetic acid (NAA) under different light conditions for 10 days to investigate effect of light on betacyanin accumulation and its relation to activity and expression of tyrosinase and expression of dopa-4,5-dioxygenase gene (DODA). Both light quality and quantity affected betacyanin synthesis of S. salsa calli. 80 lmol m -2 s -1 of white light… Show more

“…White light has been demonstrated to produce a beneficial effect on the accumulation of alkaloids in R. graveolens cultures and of flavonoids in Citrus aurantium cultures, as well as on the production of cardiac glycosides by Digitalis lanata or indole alkaloids and anthocyanin glycosides in Catharanthus roseus cultures (Ramawat and Mathur 2007). White light also stimulates the accumulation of betacyanins in callus cultures of Suaeda salsa (Zhao et al 2010).…”

“…alkaloids, anthocyanins glycosides and flavonoids (Ramawat and Mathur 2007). Also, recently the influence of light quality on the accumulation of other groups of plant pigments such as naphthoquinones and betacyanins has been studied (Zhang et al 2010;Zhao et al 2010). A few papers have dealt with the effect of light quality on the accumulation of phenolic acids, e.g.…”

“…The production of betacyanins in Sueda salsa suggests that the betacyanin pigment may function as a ROS scavenger, thus limiting the oxidative stress caused by different environmental stress factors, especially light quantity and quality (Wang et al 2007;Zhao et al 2010). Moreover, tyrosinase, which is the main enzyme in betacyanin biosynthesis, was found to be able to utilize the superoxide radical to produce melanin in human skin (Perluigi et al 2003).…”

“…Production of betacyanin -After establishment of the experiment, the calli from all callus induction media (CIM) were transferred to Betacyanin induction media (BIM) (Zhao et al 2010), consisting of MS, containing 0.3% phytagel, 30 g L -1 sucrose, 100 mg L -1 inositol, 0.5 mg L -1 adenine, 3 mg L -1 ascorbic acid and supplemented with 0.5 mg L -1 of thidiazuron (TDZ) and 1 mg L -1 naphthaleneacetic acid (NAA). Calli remained in this medium for 45 days under white light on a 16 h photoperiod at a temperature of 25 °C ± 2.…”
