Lignan Glycosides from Urena lobata

Abstract: Four new lignan glycosides; urenalignosides A–D (1–4), along with 12 known ones (5–16) were isolated from Urena lobata. Their structures were determined on the basis of extensive spectroscopic and spectrometric data (1D and 2D NMR; IR; CD; and HRESIMS). Compounds 2–4; 6; 7; 10; and 11 showed inhibition of nitric oxide production in lipopolysaccharide-induced RAW 264.7 macrophage cells with IC50 values in the range of 25.5–98.4 μM (positive control; quercetin; IC50 = 7.2 ± 0.2 μM).

“…The molecular formula C 18 H 20 N 2 O 4 was established by HRESIMS at m/z 351.1413 [M + Na] + ( Figure S37 ) and 13 C NMR data ( Figure S31 ). The 1 H NMR data of 4 ( Table 2 ) indicated the presence of an ABX pattern [ δ H 7.43 (1H, d, J = 2.0 Hz, H-2), 6.83 (1H, d, J = 8.0 Hz, H-5), and 7.41 (1H, dd, J = 2.0, 8.0 Hz, H-6)] [ 21 ], a monosubstituted benzene moiety [ δ H 7.39 (2H, m, H-3′,5′) and 7.32 (3H, m, H-2′,4′,6′)], three methylene moieties [ δ H 4.52 (2H, s, H-13), 3.92 (2H, t, J = 8.0 Hz, H-9), and 3.55 (2H, t, J = 8.0 Hz, H-10)], and a methoxy group [ δ H 3.79 (3H, s, -OCH 3 )]. The 13 C NMR spectroscopic data ( Table 2 ) of 4 displayed resonance for 16 carbons, which was confirmed by the DEPT ( Figure S32 ) and HSQC ( Figure S34 ) experiments to be a·methoxy group [ δ C 55.5 (-OCH 3 )], 3 methylenes [ δ C 49.8 (C-9), 48.2 (C-13), and 31.1 (C-10)], 6 methines, and 6 quaternary carbons.…”

“…Scientific reports indicate that the plant has anti-bacterial, antihyperglycemic, anti-nociceptive, anti-diarrheal, antiinflammatory, and wound healing activities [8,22,28]. A recent study suggests that compounds from U. lobata can inhibit nitric oxide production in lipopolysaccharide-induced RAW264.7 macrophage cells [20]. In China, U. lobata is clinically used to treat pathological leucorrhea and gonorrhea [23].…”

“…In China, U. lobata is clinically used to treat pathological leucorrhea and gonorrhea [23]. To date, a number of bioactive constituents have been isolated from this medicinal herb; these include flavonoids, phenylethyl glycosides, lignans, coumarins, triglycerides [14,20,25,35,37], megastigmane glycosides [36], urenalignosides A-D, and lignins [20].…”
