Limited diversity of T-cell receptor gamma-chain expression of murine Thy-1+ dendritic epidermal cells revealed by V gamma 3-specific monoclonal antibody.

Abstract: To study the origin of and the degree of T-cell antigen receptor (TCR) diversity of Thy-1+ dendritic epidermal cells (Thy-1+ dECs) in mice, we have developed a monoclonal antibody (mAb 536) to the yb TCR. mAb 536 binds to and stimulates interleukin 2 secretion from Thy-1+ dEC but not cells that express TCR composed of a and 13 chains. mAb 536 precipitates CD3-associated y and 6 chains from lysates of radioiodinated Thy-1+ dECs. Analysis of a panel of hybridomas that express y6 TCR indicated that mAb 536 define… Show more

“…Because fetal thymocytes expressing V␥5-bearing TCRs were previously reported to be precursors of DETCs (10,18,39,40), we first examined the development of fetal thymocytes in V␦1 Ϫ/Ϫ mice. The development of ␥␦TCR ϩ and V␥5 ϩ fetal thymocytes of V␦1 Ϫ/Ϫ mice was markedly impaired during the early-middle fetal period (GD15-GD18); thereafter, the number of ␥␦TCR ϩ , but not V␥5 ϩ , thymocytes recovered in newborn mice (Fig.…”

“…1). y and S chains of all mammary hybridomas were found to have identical apparent molecular masses and to comigrate with the receptor proteins of a V64/Vy4+, C57B1/6 day 16 fetal thymus-derived hybridoma, 33BTE-67 .1 (9) . Thus, gel electrophoresis confirmed the results of the staining experiments and suggested Vy4 JCy1 as part of the surface-expressed y/S heterodimer.…”
