Lineage determination of CD7+ CD5− CD2− and CD7+ CD5+ CD2− lymphoblasts: Studies on phenotype, genotype, and gene expression of myeloperoxidase, CD3ϵ, and CD3δ

Abstract: The gene expression of myeloperoxidase (MPO), CD3 epsilon, and CD3 delta molecules, the gene rearrangement of T-cell receptor (TCR) delta, gamma, and beta and immunoglobulin heavy (IgH) chain, and the expression of cell-surface antigens were investigated in seven cases of CD7+ CD5- CD2- and four cases of CD7+ CD5+ CD2- acute lymphoblastic leukemia or lymphoblastic lymphoma (ALL/LBL) blasts, which were negative for cytochemical myeloperoxidase (cyMPO). More mature T-lineage blasts were also investigated in a co… Show more

“…Cases 1-5 and 9-12 in this report are cases 1-5 and 8-11 in the previous report [21], respectively. This should be noted, since the results of the gene expression of myeloperoxidase, CD3e and CD36 are referred to for the consideration of the lineage of CD7+ CD5-CD2or CD7+ CD5 + CD2-blasts.…”

“…CD13 and/or CD33 antigen expression is very often present or induced in vitro in the blasts at the prothymic stage (Tables I, II), as has been shown [21]. The prolymphocytic leukemia cases in the peripheral stage T-lineage neoplasms (see Table IV) include one CD3+ CD8+ (cases 44), 2 CD3+ CD4+ (cases 41 and 48) and 2 CD3-CD4+ (cases 41 and 48) cases.…”

“…The induction of CD13 and/or CD33 has been reported in the culture of T-lineage acute lymphoblastoid leukemia cells. 35 Conversely, the antigenic loss of CD16 might occur during the culture of NK cells in the presence of IL-2. However, it was not clear in the present study whether these phenotypic changes were caused by antigenic modulation during culture or by selective growth of the CD16 − , CD33 + , CD57 − population which has an advantage in cultures containing IL-2.…”
