Lipid Nanoparticles for Organ-Specific mRNA Therapeutic Delivery

Abstract: Advances in the using in vitro transcribed (IVT) modRNA in the past two decades, especially the tremendous recent success of mRNA vaccines against SARS-CoV-2, have brought increased attention to IVT mRNA technology. Despite its well-known use in infectious disease vaccines, IVT modRNA technology is being investigated mainly in cancer immunotherapy and protein replacement therapy, with ongoing clinical trials in both areas. One of the main barriers to progressing mRNA therapeutics to the clinic is determining h… Show more

“…These large eVLPs have a size greater than 100 nm, so there is a chance that they might aggregate at the site of injection and will not reach the lymph nodes that limit their application ( Keikha et al, 2021 ). In comparison, lipid nanoparticles carrying mRNA vaccines have a size range of 100 nm or less and show high efficacy in delivering vaccines to the targeted site ( Editorial., 2021 ; Żak and Zangi, 2021 ).…”

“…Depending on the LNP composition, most ionizable lipid-based formulations have been identified from screening for liver delivery and require coating by ApoE to transfect cells in vitro, and presumably, in vivo receptor-mediated endocytosis [ 59 , 60 ]. Although they lack true tissue tropism, preferential tissue targeting and certain biodistribution profiles can be achieved by either directly engineering targeting motifs, route of administration (ROA), or altering the composition and physicochemical characteristics of the particles [ 57 , 61 , 62 ].…”

“…Preclinical studies for Onpattro® most likely used MAR, as they contain data on intracellular radioactivity in hepatocytes, hepatic vascular cells (cell type not specified), and liver sinusoid lumen [22] . Obtaining cellular resolution has a more prominent role when cell-targeting moieties are used on LNPs, for example [71] , [72] , [73] , [74] , [75] , [76] . In this regard, MAR can sometimes (depending on reagent compatibility) be coupled to immunohistochemistry to verify targeted delivery [52] .…”
