Lipid Signaling in Pathogenic Fungi

Abstract: Lipid signaling in pathogenic fungi has been studied to determine the role of these pathways in fungal biology and human infections. Owing to their unique nature, they may represent targets for future antifungal treatments. Farnesol signaling was characterized as a quorum-sensing molecule, with exposure inhibiting filamentation. Research has shown involvement in both the Ras1-adenylate cyclase and MAP kinase pathways. In species of Aspergillus, farnesol exposure induces apoptosis-like changes and alterations i… Show more

“…In general, glycosylated Cers are important signaling molecules in multicellular organisms ( 49 ), but no specifi c functions for PI-Cers are known in lower eukaryotes, except that inhibition of AUR1 is lethal to S. cerevisiae ( 50 ) and that PI-Cer is at least indirectly associated with the pathogenicity of Cryptococcus neoformans ( 51 ). A possible signaling function of PI-Cers in myxobacterial fruiting body formation or sporulation remains to be investigated.…”

“…Lipids are involved in fungal development and pathogenicity (39,100,114,127). In C. albicans, lipid molecules such as fatty acids and eicosanoids have been reported to modulate the Y-H transition.…”

“…These signaling events are stimulated by cell growth, differentiation, and external stimuli, among other signaling factors. Signaling events involved in laccase regulation include those in response to changing levels of glucose, nitrogen, oxygen, and the metal cofactors iron and copper; they also include the adenyl cyclase pathway (cyclic AMP and downstream targets), the MAPK STE12 and high osmolarity glycerol (HOG1) pathways, and sphingolipid production [34,[66][67][68][69][70][71][72].…”

“…Sphingolipid production is an important pathway for maintaining cell membrane integrity, which is critical both for growth of the fungus in vitro and for the expression and cell wall localization of laccase [20,72]. Inositolphosphoryl ceramide synthase (IPC1) is a fungal enzyme that can regulate diacylglycerol (DAG) production.…”
