Lipidomic Phenotyping Reveals Extensive Lipid Remodeling during Adipogenesis in Human Adipocytes

Abstract: Differentiation of preadipocytes into mature adipocytes is a highly complex cellular process. At lipidome level, the adipogenesis remains poorly characterized. To investigate the lipidomic changes during human adipogenesis, we used the LipidyzerTM assay, which quantified 743 lipid species from 11 classes. The undifferentiated human SGBS cell strain showed a heterogeneous lipid class composition with the most abundant classes, phosphatidylethanolamines (PE), phosphatidylcholines (PC), and sphingomyelins (SM). T… Show more

“…Lipidomic changes during the adipocyte differentiation process were also observed by Miehle et al, who followed lipidomic alterations during adipogenic differentiation of the SGBS cell line. They observed a more substantial increase in the NL fraction in 2D cell cultures compared to our primary SVF-based system, however these differences could be caused by the use of different differentiation protocols and the use of human cell lines versus primary murine cells in our system 68 . We then evaluated changes in the lipidome of the organoids by treating them with fatty acids or insulin.…”

“…Plotting the PCA and PLS-DA scores lets one visually display the classification of samples, such that the greater separation degree of the two groups, the more significant are their differences ( Miehle et al, 2020 ). Here, the results indicated the main components of lipidomics differed between the methionine-supplemented and -restricted group.…”

“…The results from our current study offer support for this paradigm. Indeed, pioglitazone caused a shortening and increased saturation of fatty acyl chains in glycerophospholipids, a trait that was recently shown to characterize the differentiation of preadipocytes into mature adipocytes, at least for in vitro models of adipogenesis ( Miehle et al, 2020 ). Moreover, pioglitazone treatment increased indices of DNL, which is another demonstrable feature of smaller, more insulin sensitive adipocytes ( Roberts et al, 2009 ).…”
