Lipoaspirate Storage Time and Temperature: Effects on Stromal Vascular Fraction Quality and Cell Composition

Abstract: The adipose tissue-derived stromal vascular fraction (SVF) is a promising candidate for use in cell therapy and tissue engineering due to its regenerative and immunomodulatory properties. Some therapies are based on using the complete SVF product, whereas others depend on the expansion of adipose-derived stromal cells (ASCs) in culture. The latter application often involves a time delay between adipose tissue harvest and SVF isolation. This study investigated how storage time and temperature affected cell qual… Show more

“…The stromal vascular fraction was harvested as previously described [ 35 ]. Briefly, lipoaspirate tissue was digested with collagenase (Nordmark, N0002880) at 37°C for 45–60 min under constant stirring.…”

“…The SVF was isolated from 4 residual lipoaspirates from plastic surgery as previously described [Svalgaard et al, 2020]. Briefly, the lipoaspirates were washed with HBSS (Gibco) and digested with collagenase (Serva/Nordmark) for 45 min at 37°C followed by inactivation with complete media (DMEM, Invitrogen, supplemented with 10% pHPL, produced in-house from a pool of 4 healthy donors) [Glovinski et al, 2017], 1% penicillin/streptomycin (Gibco), and 2 IE/mL Heparin (Amgros I/S).…”

“…Next, we prepared sets dedicated for the collection and transport of AT. ATs were transported at the temperature of 2-8 • C, which allowed us to maintain the optimal tissue quality for the isolation of SVF cells and the expansion of AT-MSCs [34].…”
