Lipoic Acid Synergizes with Antineoplastic Drugs in Colorectal Cancer by Targeting p53 for Proteasomal Degradation

Abstract: Lipoic acid (LA) is a redox-active disulphide compound, which functions as a pivotal co-factor for mitochondrial oxidative decarboxylation. LA and chemical derivatives were shown to target mitochondria in cancer cells with altered energy metabolism, thereby inducing cell death. In this study, the impact of LA on the tumor suppressor protein p53 was analyzed in various colorectal cancer (CRC) cell lines, with a focus on the mechanisms driving p53 degradation. First, LA was demonstrated to trigger the depletion … Show more

“…The antioxidant effects of ascorbic acid and liposomal ALA as an adjuvant in cancer treatment were recently re-stated [66][67][68] in ovarian carcinoma cells [69], human breast cells [70,71], non-small lung cancer cells [72], human colon cancer cells [73] or hepatoma cells [74].…”

“…4e). LA was included as a known activator of Nrf2 26 , and thus caused Nrf2 stabilization with concomitant nuclear translocation. The lack of HO-1 induction by LA despite the observed Nrf2 activation is consistent with the notion that HO-1 induction by Nrf2 requires the inactivation of the transcriptional repressor BACH-1 via heme binding 27,28 .…”

“…Upon LA treatment, the p53-independent intrinsic mitochondrial pathway was elicited in CRC cells [ 197 , 198 ]. Other effects included the induction of (macro-) autophagy, the depletion of the direct DNA damage reversal protein O 6 -methylguanine DNA-methyltransferase (MGMT) and the tumor suppressor p53 in CRC cells [ 199 , 200 ].…”
