Liposome-targeted delivery for highly potent drugs

Abstract: Considerable adverse side effects and cytotoxicity of highly potent drugs for healthy tissue require the development of novel drug delivery systems to improve pharmacokinetics and result in selective distribution of the loaded agent. The introduction of targeted liposomal formulations has provided potential solutions for improved drug delivery to cancer cells, penetrating delivery through blood-brain barrier and gene therapy. A large number of investigations have been developed over the past few decades to ove… Show more

“…Liposomes, the most widely investigated drug delivery system, have high accumulation in the liver while low immunogenicity, biocompatibility and drug protection [21,22]. Furthermore, liposomes conjugated with specific ligands could modify the pharmacokinetic characteristics and tissue distribution profile of drugs.…”

“…Liposomes were first described in 1965 and since then, they are significantly investigated and utilized as a drug carrier from last five decades. They are phospholipid vesicles made up of single or multiple concentric lipid bilayers enclosing aqueous mediums [13]. Generally, a typical liposome (50-450 nm) made up of a single bilayer lipid membrane (unilamellar liposomes) or several bilayer lipid membranes (multilamellar liposomes).…”

“…Liposomes are small spherical shape lipoid vesicles fabricated from cholesterol and various natural phospholipids. They help to improve the solubility, bioavailability, intracellular uptake of components, in vitro and in vivo stability, the pharmacokinetics and bio-distribution pattern of active moieties [25]. Biocompatibility, biodegradability, site-specificity, low toxicity and ability to entrap both hydrophobic and hydrophilic molecule are the striking features of liposomes that make them highly favoured in cosmetic and pharmaceutical industries [26].…”
