Lipoxygenase inhibitors from natural plant sources. Part 1: Medicinal plants with inhibitory activity on arachidonate 5‐lipoxygenase and 5‐lipoxygenase[sol ]cyclooxygenase

Abstract: Arachidonate 5-lipoxygenase is the key enzyme in leukotriene biosynthesis and catalyses the initial steps in the conversion of arachidonic acid to biologically active leukotrienes. Leukotrienes are considered as potent mediators of inflammatory and allergic reactions and regarding their pro-inflammatory properties the inhibition of 5-lipoxygenase pathway is considered to be interesting in the treatment of a variety of inflammatory diseases. Besides 5-lipoxygenase inhibitors, drugs able to block the 5-lipoxygen… Show more

“…Negative correlation was found between the antioxidant activities and the total phenolic and flavonoids content (Table 5). properties the inhibition of 15-lipoxygenase pathway is considered to be interesting in the treatment of a variety of inflammatory diseases (Schneider and Bucar, 2005). The IC 50 values of extracts obtained in 15-LOX inhibitory activity are similar to those of other LOX active crude extracts in the range of 1-100µg/mL (Schneider and Bucar, 2005).…”

Particularly Burkea africana with most potent activity (37.25±0.88 µg/mL) could be regarded as a potential source for new 15-LOX inhibitors.…”

“…Higher value of LOX activity IC 50 = 62.6 µg/ml with the bark of F. rhynchophylla has been found according to Huh et al (2015). Therefore, the selective inhibition of LOX is an important therapeutic strategy for asthma (Shah et al, 2011;Schneider and Bucar. 2005).…”

“…15-Lipoxygenase (15-LO) is an enzyme present in multiple systems and organs of the body that reacts with unsaturated fatty acids producing active lipid metabolites that are involved in a number of disease states such as cancer, psoriasis, atherosclerosis, and diabetes (both type 1 and 2) (Dobrian et al, 2011;Schneider & Bucar, 2005). For this reason, 15-LO has emerged as an attractive target for therapeutic intervention (Sadeghian et al, 2009).…”
