Liquid and Frozen Storage of Agouti (Dasyprocta leporina) Semen Extended with UHT Milk, Unpasteurized Coconut Water, and Pasteurized Coconut Water

Abstract: This study evaluated the effects of semen extension and storage on forward progressive motility % (FPM%) in agouti semen. Three extenders were used; sterilized whole cow's milk (UHT Milk), unpasteurized (CW) and pasteurized coconut water (PCW), and diluted to 50, 100, 150, and 200 × 106 spermatozoa/ml. Experiment 1: 200 ejaculates were extended for liquid storage at 5∘C and evaluated every day for 5 days to determine FPM% and its rate of deterioration. Experiment 2: 150 ejaculates were extended for storage as … Show more

“…Therefore, obtaining information regarding reproduction of this species will also provide information for their rational production and consequent preservation [11]. In this regard, several studies have been performed involving the reproductive characteristics of agoutis, such as placentation [10], morphology of the ovaries [12] and oviducts [13], semen collection, and semen cryopreservation [14,15].…”

“…Each aliquot of semen was slowly added by one half of experimental extender. The other half was added 20 minutes later to minimize osmotic shock (12). To minimize cold shock, those four liquid experimental semen were placed into a 20 ml beacker glass containing 50 ml water at 10 -21°C then keep it in refrigerator.…”

“…Despite this value being twice higher than that verified for ejaculated sperm (12% motile sperm) cryopreserved in UHT milk and coconut water extenders (Mollineau et al 2011), it is hypothesized that agouti presents species-specific sensitivity to the sperm freezing procedures.…”

“…Recently, the cryopreservation of agouti ejaculated sperm was reported (Mollineau et al 2011). Also, a technique to obtain epididymal sperm in this species was described (Ferraz et al 2011), but no protocol for the conservation of this valuable genetic material is available for agoutis.…”
