Liquid chromatography and capillary electrophoresis in glycomic and glycoproteomic analysis

Abstract: Glycosylation is one of the most significant and abundant post-translational modifications in cells. Glycomic and glycoproteomic analyses involve the characterization of oligosaccharides (glycans) conjugated to proteins. Glycomic and glycoproteomic analysis is highly challenging because of the large diversity of structures, low abundance, site-specific heterogeneity, and poor ionization efficiency of glycans and glycopeptides in mass spectrometry (MS). MS is a key tool for characterization of glycans and glyco… Show more

“…When saccharides are not separated with HILIC, for example, isomers [176], RP-LC and derivatization with PMP may solve the problems of separation performance, sensitivity, and repeatability for monosaccharides [179]. Furthermore, monolithic nano-LC and HILIC coupled with MS give efficiency in glycaemic and glycoproteic studies [181], and accuracy in the identification of structural functionalities. However, drawbacks appear due to sample preparation, since low-molecular saccharides may not be origin from poly-or oligosaccharides when their fragments are detected as monosaccharides [177,179].…”

“…CZE is especially useful for the characterization of PTMs that influence the charge of peptides or proteins. Characterization of glycosylation of proteins in their intact form by different CE and LC methods has been recently reviewed [138,139].…”

“…17 The available techniques for quantifying ATZ in plasma are currently limited. These techniques include liquid chromatography-tandem mass spectrometry (LC-MS/MS) [18][19][20] and enzyme-linked immunosorbent assays (ELISAs). 21,22 However, LC-MS/MS has several signicant drawbacks.…”
