Liquid chromatography–atmospheric pressure chemical ionization tandem mass spectrometry for the simultaneous determination of dimethoxyaschantin, dimethylliroresinol, dimethylpinoresinol, epimagnolin A, fargesin and magnolin in rat plasma

Abstract: A rapid, selective, and sensitive liquid chromatography-atmospheric pressure chemical ionization (APCI) tandem mass spectrometry method was developed for the simultaneous determination of dimethoxyaschantin, dimethylliroresinol, dimethylpinoresinol, epimagnolin A, fargesin and magnolin, the pharmacologically active ingredients of Magnolia fargesii in rat plasma. These tetrahydrofurofuranoid lignans were extracted from rat plasma using tert-butyl methyl ether at pH 7.4. The analytes were separated on a Pinnacle… Show more

“…Epimagnolin, a natural compound extracted from Magnolia flos (Shin‐Yi), which is a traditionally used oriental medicinal herb, is a major ingredient used to treat nasal congestion, sinusitis and anti‐inflammation . Epimagnolin has a chemical structure similar to those of magnolin and aschantin . Recently, our research group found that magnolin targets ERK1 and ERK2 with 87 and 16.5 nM IC 50 values, respectively .…”

“…However, typical pharmacokinetic studies can be difficult to apply to herbal drugs that consist of multiple constituents, with frequently unidentified pharmacologically active components. As such, simultaneous determination of the various potential active constituents, major constituents, and/or their metabolites is recommended for the satisfactory pharmacokinetic evaluation of herbal drugs [ 21 , 22 , 23 ].…”

“…Magnolin also ameliorates contrast-induced nephropathy in rats via anti-oxidation and anti-apoptosis [17]. The ethanol extract of the dried flower buds of M. fargesii (encoded as NDC-052 contained eudesmin, fargesin, epimagnolin A, magnolin, and yangambin as 4.1, 3.4, 11.9, 21.5, and 9.1%, respectively, as determined by the LC-APCI-MS/MS method [18]) has been developed as an effective alternative or complement to standard asthma therapy based on their biological activities [19]. Consequently, taking NDC-052 (600 mg/day for 8 weeks per oral) in adult asthmatic patients was safe and tolerated [19].…”
