Live‐Cell Imaging and Analysis with Multiple Genetically Encoded Reporters

Abstract: Genetically encoded live-cell reporters measure signaling pathway activity at the cellular level with high temporal resolution, often revealing a high degree of cell-to-cell heterogeneity. By using multiple spectrally distinct reporters within the same cell, signal transmission from one node to another within a signaling pathway can be analyzed to quantify factors such as signaling efficiency and delay. With other reporter configurations, correlation between different signaling pathways can be quantified. Such… Show more

“…At the same time, the authors monitored the activation of ERK by the nuclear translocation of a fluorescently tagged ERK; the transcription of target genes using an engineered sequence within the nascent RNA that induces the localisation in sites of transcription of a second fluorescent protein; and the expression of the target protein tagged with a third genetically encoded fluorophore. Other innovations in fluorescence microscopy, such as high-content imaging, multiplexed biochemical imaging are also laying the foundation for systems level understanding of cell decisions [ 34 , 55 , 73 , 74 , 75 , 76 , 77• , 78 ].…”

“…Each imaging dataset presented is representative of at least two independent replicate experiments. Image, data processing, and statistics were performed using custom MATLAB software as previously described (Pargett and Albeck, 2018;Pargett et al, 2017). Linear regression was performed using a Theil-Sen estimator as previously described (Gillies et al, 2017).…”
