Live Cell Imaging of Male Meiosis in Arabidopsis by a Landmark-based System

Abstract: Live cell imaging has tremendously promoted our understanding of cellular and subcellular processes such as cell division. Here, we present a step-by-step protocol for a robust and easy-to-use live cell imaging approach to study male meiosis in the plant Arabidopsis thaliana as recently established. Our method relies on the concomitant analysis of two reporter genes that highlight chromosome configurations and microtubule dynamics. In combination, these reporter genes allowed the discrimination of five cellula… Show more

“…Different time points of fixation were carried out (12, 28, and 38 h). Given the known meiotic timecourse of A. thaliana [16][17][18] (Figure 3a), the different time points and inhibition methods could provide us with insights about at what stage of meiosis TOPII may be functional. The treatment with merbarone (Figure 3b-e) produced the presence of multiple ABs at AI (6% n = 50) (Figure 3b), chromosome fragmentation (7% n = 43) (Figure 3c), missegregation (4% n = 25) (Figure 3d), and micronuclei at telophase II (10% n = 40) (Figure 3e).…”

“…The meiotic time-course in A. thaliana has been previously characterized by using the nucleotide analogue BrdU [16,17]. This time course has been validated recently with life cell imaging techniques [18].…”

“…Live-cell imaging techniques have been recently applied to A. thaliana meiosis but, until now, have mainly been used to study meiotic chromosome segregation 32 34 . Here, we applied the approach described by ref.…”

“…There is no precise description in plants of the RPMs nor of any machinery components involved in chromosome movement. We took the opportunity of recent advances in live imaging of meiosis in A. thaliana 32 34 to develop an approach to analyze meiotic prophase centromere dynamics in this model plant. Using 3D fluorescence time-lapse microscopy on male reproductive organs (anthers) and quantitative image analyses, we describe for the first time A. thaliana RPMs.…”
