LncRNA LINC-PINT Inhibits Malignant Behaviors of Laryngeal Squamous Cell Carcinoma Cells via Inhibiting ZEB1

Abstract: Objective: Laryngeal squamous cell carcinoma (LSCC) belongs to head and neck squamous cell carcinoma (HNSCC), with dismal prognosis. Here, this study aims to disclose the role of LINC-PINT in cancer development, which may contribute to improving the clinical outcomes of LSCC treatment. Methods: LINC-PINT expression in LSCC tissues and in TU-177 and Hep-2 cells was quantified, and subsequently, the association between LINC-PINT and LSCC malignancies was analyzed. pcDNA3.1-LINC-PINT or pcDNA3.1-EZH2 was introduce… Show more

“…ZEB1 accumulation enhanced the invasion and EMT of liver and gastric cancer cells (36,37). Importantly, ZEB1 acts as an oncogene and is closely related to EMT and prognosis in LSCC (38,39). Our study shows that ZEB1 expression was enhanced in LSCC cells, and it was inhibited by silencing HCP5 and promoted by cosilencing HCP5 and miR-216a-5p.…”

“…We analyzed each gene in the risk signature, NR2F2-AS1 induced epithelial-mesenchymal transition in non-small cell lung cancer, promoted prostate cancer cell proliferation by regulating CDK4, and modulated the MAPK pathway to regulate colorectal cancer invasion and metastasis [30][31][32]. LINC-PINT inhibited the proliferation and migration of laryngeal squamous cell carcinoma by silencing ZEB1 [33],…”

“…Here, LINC‐PINT can directly interact with EZH2 to enhance H3K27me3‐mediated repression of key genes involved in melanoma progression including CDK1, CCNA2, AURKA, and PCNA (Xu et al, 2019), thus inhibiting the proliferation of melanoma cells. In similar fashion, LINC‐PINT also enriches EZH2 to silence ZEB1 which inhibits the proliferation, migration, and invasion capabilities of laryngeal squamous cell carcinoma cells (Yang et al, 2021). Thus, a common theme is emerging that LINC‐PINT function by recruiting EZH2 to target gene promoters, leading to H3K27 trimethylation and silencing of the genes important for promoting tumorigenicity (Figure 2a).…”

“…Restoring LINC‐PINT expression in LSCC cell lines inhibits their proliferation, migration and invasion through the silencing of ZEB1 which involves the activation of EZH2 (Table 1). The levels of phosphorylated AKT/mTOR also declined upon LINC‐PINT overexpression (Yang et al, 2021). The identification of EZH2 involvement is a familiar theme for LINC‐PINT but the independent biological function exerted through AKT/mTOR underscores the multifaceted nature of LINC‐PINT's actions in cancer.…”
