Localisation of aphidicolin-induced break points in Holstein-Friesian cattle (Bos taurus) using RBG-banding

Abstract: Fragile sites (FS) seem to play a role in genome instability and may be involved in karyotype evolution and chromosome aberrations. The majority of common fragile sites are induced by aphidicolin. Aphidicolin was used at two different concentrations (0.15 and 0.30 μM) to study the occurrence of FS in the cattle karyotype. In this paper, a map of aphidicolin induced break points and fragile sites in cattle chromosomes was constructed. The statistical analysis indicated that any band with three or more breaks wa… Show more

“…FS-inducing agents include mutagenic and clastogenic chemicals [58]. Fragile sites must be induced with chemical compounds because the spontaneous expression of fragile sites common on metaphase plates is very low (less than 5%), which makes them very difficult to detect [59]. Fragile sites are a natural element in the chromosome structure of all species and show genetic conservatism in the evolution of mammals [60].…”

“…Various frequencies of damage within a species have been observed by researchers analysing karyotypes of various breeds of animals (Di Berardino et al 1983;Iannuzzi et al 1991;Riggs et al 1993;Ronne 1995;Di Meo et al 2000;Rodriguez et al 2002;Danielak-Czech and Słota 2004;Peretti et al 2007Peretti et al , 2008Nicodemo et al 2008;Albarella et al 2009;Wójcik et al 2011Wójcik et al , 2012. Differences between mean numbers of various forms of damages in genetic material result from the fact that their frequency is influenced by the breed.…”

“…1996). In cattle, various fragile sites were identified (Rodriguez et al. 2002); thus, we must consider the eventuality that these sites are more frequently involved in RCP formation.…”

“…In humans, chromosomes carrying fragile site show more inclination to be involved in RCP than expected (Cohen et al 1996). In cattle, various fragile sites were identified (Rodriguez et al 2002); thus, we must consider the eventuality that these sites are more frequently involved in RCP formation. Moreover, if we presume the existence of an RCP deriving from two fragile site breakpoints, the likelihood of being visible is very low.…”
