Locally Controlled Release of Methotrexate and Alendronate by Thermo-Sensitive Hydrogels for Synergistic Inhibition of Osteosarcoma Progression

Abstract: Osteosarcoma (OS) is a serious primary bone malignant tumor that can easily affect children and adolescents. Chemotherapy is one of the important and feasible clinical treatment strategies for the treatment of OS at present, which is severely limited due to insufficient retention time, poor penetration ability, and serious side effects of current antitumor drug preparations. In this work, a novel injectable thermo-sensitive hydrogel (mPEG 45-PLV 19) loaded with methotrexate and alendronate, and the sustained r… Show more

“…The aforementioned strategy has been followed, wherein injectable thermo-sensitive poly (L-valine) hydrogel containing methotrexate and alendronate was employed in osteosarcoma progression, which managed sustainable release at the tumor site. More precisely, this developed system has reduced bone destruction and lung metastasis due to osteosarcoma (reduction in bone density, hence weakness) [ 108 , 109 ].…”

“…However, some complications in preparation and applications are also associated with the hydrogels. Therefore, the specific site of application and its clinical effectiveness still need to be explored [ 109 ].…”

“…Hence, several scientists developed MTX loaded hydrogel to improve solubility and lower the side effects and treat cancer [ 61 , 130 ]. For example, a novel MTX and alendronate co-loaded mPEG45–PLV19 containing thermosensitive hydrogel was developed by scientists, demonstrating synergistic inhibition osteosarcoma with sustained release of drugs [ 131 ]. In another study, investigators developed in situ MTX loaded gelatin and poly(vinyl) alcohol (Gel/PVA) hydrogel and used to treat colorectal disorders [ 132 ].…”

“…mPEG 45 -PLV 19 (5.0 wt%) was stirred slowly and thoroughly at 4 • C overnight, and OXA (5.0 mg/mL) (Feather et al, 2018) and ALN (10.0 mg/mL) (Shan et al, 2020) were added to the polymer solution and mixed thoroughly (4 • C) to keep injectable. The sol state is used for tumor suppression experiments.…”
