lon transcriptional regulation of genes necessary for capsular polysaccharide synthesis in Escherichia coli K-12

Abstract: It has previously been observed that Escherichia coli lon mutations increase the levels of enzymes involved in the synthesis of colanic acid capsular polysaccharide (A. Markovitz, in I. Sutherland, ed., Surface Carbohydrates of the Prokaryotic Cell, 1977). To determine how lon regulates these enzymes, we have isolated, mapped, and characterized lac operon and lac protein fusions to genes necessary for capsule synthesis by the Mu d(lac Amp) in vivo fusion technique of Casadaban and Cohen (M. J. Casadaban and … Show more

“…However, as others have observed (Holder et al, 1985), some of them are subject to proteolysis. In our hands, proteasedeficient strains (Trisler and Gottesmann, 1984) did not significantly improve the expression yield or stability of these proteins.…”

“…For DNA transformations the E. coli K12 strain HB 101 was used. Recombinant proteins were expressed in E. coli strains M15 (Certa et al, 1986) or SG 200-50 (Ion A-; Trisler and Gottesmann, 1984).…”

“…To clarify this role, two mutants were constructed: (i) a mutant with abolished CA synthesis was constructed by transduction of a knock-out mutation in the cpsE (i.e. wcaE) gene resulting in the disruption of CA operon transcription (Trisler and Gottesman, 1984;Stevenson et al, 1996), and (ii) a mutant overproducing CA, obtained by deletion of the lon gene (Trisler and Gottesman, 1984). PHL644 and these two strains were grown on M63 glucose agar plates to verify their CA production.…”

“…Briefly, at least six genes required for the synthesis of colanic acid {ops) are controlled by tv^fo negative regulators, lon and rcsC, and two positive regulators, rcsA and rcsB {Gottesman and Zipser. 1978: Gottesman et al, 1985;Trisler and Gottesman, 1984). rcsB controls the transcription of the structural genes involved in the biosynthesis of colanic acid.…”
