Long term worsening of Alzheimer pathology and clinical outcome by a single inoculation of mutated beta-amyloid seeds

Abstract: Background Alzheimer’s disease (AD) is characterized by intracerebral accumulation of abnormal proteinaceous assemblies made of amyloid-β (Aß) peptides or tau proteins. These lesions induce synaptic dysfunctions that are strongly correlated with cognitive decline. Intracerebral infusion of well-defined Aβ seeds from synthetic or recombinant non-mutated Aβ1−40 or Aβ1−42 peptides can increase Aβ plaque depositions several months after the infusion. Familial forms of AD are associated with mutations in the amylo… Show more

“…Neuroimaging studies resting state functional MRI (rs-fMRI)-blood oxygen level dependent (BOLD) sequence for brain imaging was processed using the seed-based analysis (SBA) method to analyse functional connectivity (FC) (31). SBA can evaluate functional changes between participants, analyse a region of interest (ROI), and show the FC among different brain networks such as default mode network (DMN) (32, 33), sensorimotor network (SEN) (34), executive control network (ECN) (35), language network (LN) (36), visuospatial network (VN) (37), and salience network (SAN) (38).…”
