Longitudinal analysis of serum oxylipin profile as a novel descriptor of the inflammatory response to surgery

Abstract: BackgroundOxylipins are potent lipid mediators demonstrated to initiate and regulate inflammation yet little is known regarding their involvement in the response to surgical trauma. As key modulators of the inflammatory response, oxylipins have the potential to provide novel insights into the physiological response to surgery and the pathophysiology of post-operative complications. We aimed to investigate the effects of major surgery on longitudinal oxylipin profile.MethodsAdults patients undergoing elective l… Show more

“…Over the course of ICU stay, levels of 5,6-DHET and 8,9-DHET increased in both those with brain injuries and those with pneumonia, which mirrors findings seen between the early and late post-operative periods following surgery 32 , which may reflect the anti-inflammatory properties of the precursor EETs and their role modulating pulmonary inflammation 33 . Patients who developed VAP also showed a trend to increased levels of the pro-inflammatory mediators IL-6, IL-8 and ICAM-1, reflecting an inflammatory response to infection.…”

“…Likewise, LOX-derived oxylipids display a wide range of biological actions. For instance, 15-HETE has been associated with anti-inflammatory effects, whereas 9and 13-HODE have been associated with anti-thrombotic effects in humans (Wolfer et al, 2017). In accordance with a study performed by Raphael et al (2014), plasma concentrations of 9-and 13-HODE followed a similar trend in healthy cows across the sample period.…”

“…Arachidonic acid is a major polyunsaturated fatty acid, from which some inflammatory mediators are derived in mammals ( Yagami et al., 2018 ). Arachidonic acid can be metabolized by lipoxygenases (LOX) ( Lehmann et al., 2015 ) to leukotrienes (LT), or by cyclooxygenases (COX) ( Willenberg et al., 2015 ) to prostaglandins (PGs) and thromboxanes (TXs), or by cytochrome P450s (CYP450) and LOX ( Fleming, 2011 ) to hydroxyeicosatetraenoic acids (HETEs) and hydroxyoctadecadienoic acids (HODEs) ( Wolfer et al., 2017 ). Development and progression of AS is a complicated process regulated by many internal factors, including arachidonic acid-derived lipid mediators.…”
