Loop-Mediated Isothermal Amplification Assay for Identifying Neisseria gonorrhoeae Nonmosaic penA- Targeting Strains Potentially Eradicable by Cefixime

Abstract: Neisseria gonorrhoeae is among the hot topics of “resistance guided therapy,” one of the top 5 urgent antimicrobial threats according to the Centers for Disease Control and Prevention (CDC). There is a need either to develop new agents or to make effective use of existing agents, with the current limited number of therapeutic agents available.

“…The optimum reaction conditions are 2 h at 37 °C with near-neutral medium. 19 An excellent linear correlation between bacterial concentration and the absorbance at 450 nm was further confirmed in this work (Fig. S1 †).…”

“…The loop-mediated isothermal amplification (LAMP) tests demonstrated high sensitivity and specificity for the identification of species and resistance genes in bacteria, which was conducive to guiding clinical medication. 18,19 LAMP tests neither need much equipment nor training. However, LAMP test is a genotypic category.…”

“…PBP2 is a key player in antibiotic resistance and does not exist in susceptible strains. 18 Hence, the use of LAMP assays has shown better diagnostic performance in gonorrhoea and, in the future, can be used for the identification of resistance as well.…”
