Lophodermium pini-mugonis sp. nov. on needles of Pinus mugo from the Alps based on morphological and molecular data

Abstract: Lophodermium pini-mugonis, collected on needles of Pinus mugo from German Alps, is described as a species new to science. It is characterized by subcuticular ascomata with a wrinkled surface and a somewhat untidy outline, a complex structure of lip cells, and ellipsoidal conidia. An analysis of the internal transcribed spaces of rDNA showed that Lophodermium pini-mugonis is, sister to Lophodermium autumnale and distantly related to other Lophodermium species on pines. The hypothesis of cospeciation of Lophoder… Show more

“…rather than Abies spp. The Rhytismatalean species on Abies and Picea are distally related to species on Pinus (Ortiz- García et al 2003;Hou et al 2009). Therefore, we provisionally place irula exigua in Lirula and its systematic position needs to be confirmed by more molecular data of members in the Rhytismatales on needles of Abies and Picea.…”

“…Total genomic DNA was extracted from ascomata followed Hou et al (2009). Seven new sequences for ITS rDNA and part LSU rDNA regions respectively were obtained for species of Lirula macrospora, Lirula yunnanensis, Lirula exigua, Lophodermium autumnale (Darker) Magasi, and Lophodermium piceae (Fuckel) Höhn.…”

“…Gelatinous sheaths surrounding ascospores and paraphyses were observed in water or 0.1 % (w/v) cotton blue in lactic acid. Measurements and drawing of asci, ascospores and paraphyses were made from 30 ascospores, asci, and paraphyses for each specimen using material mounted in 5 % KOH solution or Melzer's reagent (Hou et al 2009). The specimens are deposited in the Herbarium of the College of Life Science, Capital Normal University (BJTC).…”

“…Total genomic DNA was extracted from ascomata following the protocol of Hou et al (2009). New sequences for the nuclear large subunit ribosomal DNA (LSU rDNA), internal transcriber spacer (ITS rDNA) and mitochondrial small subunit ribosomal DNA (mtSSU rDNA) were obtained from the new species (HOU327) and Lophodermium camelliicola (L0447).…”
