Low Collagenase-1 (MMP-1) and MT1-MMP Expression Levels Are Favourable Survival Markers in Advanced Colorectal Carcinoma

Abstract: Objective: Extracellular matrix degradation is required for invasive growth and metastasis formation in colorectal carcinoma; therefore, we examined matrix metalloproteinases expression (MMP-1, MMP-13 and MT1-MMP) and apoptosis in tumours from 49 patients with advanced colorectal disease. Methods: MMP expression was determined immunohistochemically and apoptotic index (AI) was ascertained using the TUNEL assay. Results: Low levels of MT1-MMP, MMP-1 and AI were found to be favourable markers significantly assoc… Show more

“…Likewise, weak MMP-1 expression has been related to a better prognosis of patients with advanced CRC. 44 However, also our colon cancer patients with high MMP-1 expression showed a tendency towards worse prognosis. Expression of MMP-13 has been associated with poor prognosis in breast cancer 45 and CRC.…”

“…That MMP-1 inhibition increases the rate of apoptosis may explain previous reports that blocking of MMP-1 activity by TIMP-1 inhibits apoptosis, 38 that increased expression of this enzyme leads to tumor progression and metastasis, 34 and that low levels of MMP-1 are favorable markers for tumor spreading. 39 The rate of apoptosis in a tumor determines its growth, invasion, and metastasis and in nearly all instances a significant correlation between expression of MMP-1 and cancer survival has been documented. 34,39 -41 Because control of cell survival may be achieved through inhibition of apoptosis, it is possible that this is another mechanism by which MMP-1 promotes cell invasion and growth in inflammatory, angiogenic, and tumoral processes.…”

“…Low expression levels of MT1-MMP are considered as favorable survival markers in advanced colorectal carcinoma (Bendardaf et al, 2003). MT1-MMP has been detected in tumor cells and adjacent stromal cells in a variety of human tumors, including breast, cervical, colon, bladder, gastric, glioma, pancreatic, liver, ovarian, prostate and thyroid cancer Zucker et al, 2003).…”
