Low density lipoprotein receptor-related protein/alpha 2-macroglobulin receptor is an hepatic receptor for tissue-type plasminogen activator.

Abstract: Tissue-type plasminogen activator (t-PA), a serine protease that catalyzes the initial and rate-limiting step in the fibrinolytic cascade, is cleared rapidly in vivo by the liver. Using chemical crosslinking, we have recently identified a plasminogen-activator inhibitor type 1 (PAI-1)-independent t-PA clearance receptor on rat hepatoma MH1C1 cells with a relative molecular mass of %-500 kDa. Another recently identified membrane receptor, low density lipoprotein receptorrelated protein/a2-macroglobulin receptor… Show more

“…We selected α 2 M* for these control experiments because the binding affinity of α 2 M* for LRP1 is much higher than the affinity of tPA for LRP1 (Ney et al, 1985;Hussaini et al, 1990;Bu et al, 1992). Although ERK1/2 was robustly activated by α 2 M* in the absence of RAP through the entire 60 min timecourse, when RAP was present, p-ERK1/2 was not observed (Fig.…”

“…LRP is a unique cell surface receptor and a member of the LDL receptor gene family. LRP recognizes multiple ligands including apolipoprotein E-enriched ␤-migrating very low-density lipoprotein and methylamine-activated ␣ 2 -macroglobulin (21,22) in addition to t-PA. Cell-mediated endocytosis of t-PA and other LRP ligands can be inhibited by a 39-kDa receptorassociated protein (RAP) that copurifies with LRP (18). Although RAP has been recently shown to be an intracellular chaperone that modulates the activity of LRP along the secretory pathway (23), this molecule has been used experimentally as an effective antagonist to regulate ligand-LRP interactions on the cell surface (24).…”

“…A 39-kDa protein, termed the receptor-associated protein (RAP) (17), binds to several members of the LDL receptor family (18 -20) and blocks their ligand binding capacity. LRP mediates the cellular uptake and subsequent degradation of proteinases, such as tissue-type plasminogen activator (21) and urinary-type plasminogen activator (22), proteinase-inhibitor complexes, such as ␣ 2 -macroglobulin-proteinase complexes (23,24), soluble amyloid precursor protein (25), and apolipoprotein E-enriched lipoproteins (26,27). LRP is expressed in many tissues, and is a major endocytic receptor in the liver and in the central nervous system (for reviews, see Refs.…”
