Low doses of ipsapirone increase growth hormone but not oxytocin secretion in normal male and female subjects

Abstract: The effects of gender and age on the degree of stimulation of GH secretion by 5-HT1A agonists in human subjects differ from their effects on secretion of the hormones ACTH and cortisol. A higher dose of ipsapirone is required to stimulate oxytocin secretion in normal human subjects.

“…The very significant increase in GH after flesinoxan completely parallels those reported by Seletti et al (1995) and is consistent with studies using other 5-HT 1A agonists such as ipsapirone, gepirone or tandospirone to stimulate GH response (Lesch et al 1989;Anderson et al 1990;Miller et al 1990;Cleare et al 1998;Newman et al 1999;Cleare and Bond 2000). This hormonal effect appears to be mediated through the stimulation of post-synaptic 5-HT 1A receptors (Seletti et al 1995).…”

“…Stimulation of the hypothalamo-pituitary-adrenal axis (HPA) by the 5-HT 1A agonist ipsapirone has been demonstrated in several studies (Kahn et al 1994;Meltzer and Maes 1995a;Gelfin et al 1995;Cleare et al 1998;Schwartz et al 1999). Ipsapirone also stimulates the hypothalamic-GH axis (Lesch et al 1989;Cleare et al 1998;Newman et al 1999;Cleare and Bond 2000), as does tandospirone (Miller et al 1990) and gepirone (Anderson et al 1990). However, these 5-HT 1A compounds are partial agonists that may have agonist or antagonist activities and most of them are metabolized into 1-phenylpiperazine (1-PP), an alpha 2 -adrenoreceptor antagonist.…”

“…It appears that these binding differences have functional consequences. Ipsapirone, a 5-HT 1A partial agonist, stimulates GH secretion more in male than in female subjects indicating greater responsivity in females (Newman et al, 1999). Animal studies suggest different 5-HT 1A responses in males and females (Gelfin et al, 1995;Ebenezer and Tite, 1994;Haleem et al, 1989Haleem et al, , 1990Joppa et al, 1997;Uphouse et al, 1991;Mendelson and McEwen, 1991;Curzon, 1989).…”

“…This innervation from the PVN onto dopaminergic neurons is important for social reward [ 10,11 ]. Similarly, there is evidence that vasopressin interacts with the dopaminergic system in the lateral septum [ 12,13 ] to moderate social play in a sex-speci c manner [ 14 ]. Therefore, this raises the possibility that the oxytocin and vasopressin systems might be involved in the behavioural response to tickling, in a sexually dimorphic manner.…”
