Low Molecular Weight pDMAEMA-block-pHEMA Block-Copolymers Synthesized via RAFT-Polymerization: Potential Non-Viral Gene Delivery Agents?

Abstract: Abstract:The aim of this study was to investigate non-viral pDNA carriers based on diblock-copolymers consisting of poly(2-(dimethyl amino)ethyl methacrylate) (pDMAEMA) and poly(2-hydroxyethyl methacrylate) (pHEMA). Specifically the block-lengths and molecular weights were varied to determine the minimal requirements for transfection. Such vectors should allow better transfection at acceptable toxicity levels and the entire diblock-copolymer should be suitable for renal clearance. For this purpose, a library o… Show more

“…poly(DMAEMA)-b-poly(HEMA)-dithiobenzoate were shown to have low cytotoxicity with relation to block copolymers based on polyethyleneimine. [217] poly(DMAEA)-trithiocarbonate was shown to have low cytotoxicity and to degrade under physiological conditions to poly(AA). [444] Polymerization Kinetics Kinetic simulation is frequently used as a tool to correlate experimental data with theoretical models.…”

“…7,85 However, this can be also used to prevent non-specific interactions and allow specific internalisation due to targeting moieties at the same time, in particular in vivo. A frequently used approach is the combination of a PEG block and a cationic block, resulting in reduced cytotoxicity and stealth behaviour.…”

“…There are multitude studies about PDMAEMAbased stars. On one hand, a higher cytotoxicity was observed when keeping the arm length constant and increasing the number of arms (4,7,14,21), thus, also increasing the molar mass. In particular, T. K. Georgiou demonstrated the benefit of PDMAEMA stars as transfection agents.…”

“…Residual 30% of polymer chains are initiated by AIBN forming a mixture of homo-, diblock-and even triblock copolymers. Purification techniques like precipitation or dialysis do not always help to separate diblock copolymers from homopolymers [44]. Moreover, cleavage of Z groups sometimes takes place during purification by dialysis [45].…”
